Ok, so after a quick re-watch of the scene - Dean parks the Impala in front of the building that has a cross on it, with Jesus Saves written within it. And the marquee reads "Jesus Saves(,) All Are Welcome".

So again, that's another reason I read that upcoming scene as the woman mistaking Sam for Jesus the way I did.
