I think you're completely correct. If it were the only reference, I'd think it might be a coincidence, but it wasn't the only reference to Dean Cain having played Supes. There were a couple other lines:
Knox: You may have powers beyond those of mortal men, but this time you've met your match.
Later:
Clark: We're not as different as you think.
Both lines were during the Clark/Knox scuffle at the end of the episode. There may have been other references as well.

Didja also notice at the end when Dean Cain himself did the promo for the Christopher Reeve memorial fund? I thought that was sort of sweet seeing as how they both played essentially the very same character down to the blue tights and red cape!

I agree it was done on purpose. they even showed Knox's handkercheif with his initials CK on it.
I think they were also trying to further showcase and stress one of Clark's greatest fears; that of being alone and what it could do to him. Seeing Knox was like getting a preview of how maddening his life might become, after he "outlived every women he's ever loved" It also mirrors Chloe's similar concerns and fears
I thought that was a subtle as a trout slap. Pretty obviously a reference to how tough it'll be for Clark.

Wonderful. I know they did it on purpose. I loved how Clark saw himself in the Psychopathic Knox.

I thought it was very nice that Dean Cain had CK inital's on his handkerchief. A nice nod to Dean's work as Clark kent.

Yeah, I picked up the reference. I thought it was a neat little reference. As well as the other comments mentioned.
Also the fact that he's immortal, as Clark will wind up being pretty much immortal too.

Yeah, the CK initials handkerchief was a cool little nod. Especially since on LnC Jimmy always used to call Clark "CK".
And the thng about Clark's fear of being alone, I notice they're referring to that a lot this season. Both Kara and MM have talked about it recently and you even when Lana was talking about "growing old together" you could see on Clark's face that he was thinking about this fear. This all goes back to Cassandra's vision in Season 1 of Clark in a cemetary surrounded by the graves of everyone he loves. It's always been one of Superman's greatest worries in the mythos, and I think it's good to see Smallville is referencing that.

They were referring to the fact that Dean Cain played the role of Clark/Supes in Lois & Clark: The Adventures of Superman.

No he isnt he's just a made up character, alot of people thought he would be Vandal Savage a real character from the comics. But Knox is someone that was supposed to show how Clark could potentially be when he outlives everyone he's gotten to love and know seeing as he is immortal until the yellow sun is gone.
The coincedence is that Dean Cain (the guy play Dr. Knox) played Superman/ Clark Kent in the 1990's Abc hit show Lois and Clark the new adventures of superman. So them being similar was right on the money. though TW will never don the suit. :(

I love superman and the mythos and I was kind of confused about -besides Dean Cain's link to superman- the importance people were giving to the character of Curtis Knox, but I guess you're right and it represents what might happen to Kal/Clark in many years from now, and that he has to remember not to go evil after everyone he loves has gone. how sad....:(
ps. i loved "the new adventures of superman" I used to watch it all the time, nice to see Dean cain back BTW,

eh, he wont. thats the thing. in the comic kingdom come, Clark actually leaves metropolis after Lois dies and just stays in the fortress in some kind of hologram simulator that makes he think he's living on the farm back in smallville. he doesnt go nuts while trying to find ways to bring back his wife.
and yeah it was awesome seeing dean cain as a villian in the superman universe. his interaction with lex in the mansion was my favorite scene

No, but he shared his initials (CK) with Clark Kent. Dean Cain (the actor who played Curtis Knox) played Clark Kent on Lois & Clark in the mid 90's.
