This line has probably been said more than once: "Are you kidding"?
I think it has been said quite a few times. How about if we narrow it down to the one you are looking for? Was it said by a main or a guest? To a main or a guest?

A main to a main
Well, that rules out about half of them...
Let's see... Is it:
Chloe to Lana in Prodigal when Lana wonders if she should hire Clark to work part time at the Talon. Chloe continues, "Bossing around Clark is the highlight of my day! Besides, he always comes through, even if it is in his own special Clark time."
Lois to Clark in Kara after he tells her he is leaving Smallville after Lana's funeral and seems surprised that she is going to miss him. Lois goes on, "Who am I gonna pick on?" (probably not the one you are looking for)
Chloe to Clark, in Prodigal again, when he is surprised she knows about Lucas. She explains, "The prodigal Luthor son returns, and to top it all off, the new heir has taken over the castle? I mean, that's a story that could land you a byline on the front page of the Daily Planet."
Or Lois to Clark in Facade when he is trying to keep her out of the boys' locker room. She goes on, "Tortured senior can't keep up the hometown hero act, and buckles? This is as juicy as my teen cosmetic surgery article."

The 1st instance was in Clark's 1st dream in Slumber. At the mansion Lex asked Clark to help him with a problem.
Clark: What do you need?
Next line...
Lex: "Would you hand me that sword?"
And Lex, after chat about Katai warriors, broke the sword over Clark's forearm proving he was more than he claimed to be.
2nd instance was Lex to Isobel-in-Lana at the mansion in Spell.
Lana: "Oh, a good bottle of wine for Chloe's birthday."
Shortly after she got him to play the piano infinitas.
3rd instance was Chloe to Lana at the DP in Rage when Lana responded "I'm curious about this halfway house that LuthorCorp Foundation supports. I offered to get involved, but Lex won't let me anywhere near it. He says it isn't safe."
The 4th instance is Clark to Lois in the barn in Prototype. Lois responds "Your mom's political influence. Look, do you think that you could use your connections to get a look at Wes' military jacket?"
The 5th instance is Jimmy to Lex at Lex's DP office in Arctic after Lex had cleard Chloe's name with the DDS. Lex responded "Lois off my back. She's been asking too many people too many questions."
The 6th instance was Clark on the phone to Lois when she needed some wardrobe assistance in Warrior. Clark sped to Smallville and Lois responded "What was that whooshing sound?"

Hehe, I was going for the last one.
I think yours was Chloe to Lois at the Talon appartment in Phantom, after she convinced Lois not to go after Lex and Lois tore the piece of paper with the numbers she wrote.
The next line was after Chloe left the epartment and Lois had picked up the piece of paper and figured out what they were. She said :"Longitude and latitude. No duh"
Who said: "Maybe for a minute"?

I'm not sure if you're referring to "Oh, I think for a minute" that Martha said to Lois in Crusade since the line isn't the same as "Maybe for a minute". But if that's the one, then I think the following line was "It's funny, I never thought she'd fall for the farm boy type".

That was Clark to Chloe the 2nd time we see the mansion cellar rescue scene in Promise. It was from Lana's POV after Clark had said he was protecting her and Chloe said Lana was marrying a monster. Chloe replied "I think it's easier than getting hurt. Clark, you don't hesitate to run into a burning building or jump in front of a bullet because nothing can penetrate that iron flesh of yours. I believe that was Chloe to Clark in Sneeze. They were in Pontius's office, and he had spotted a room behind the wall, but was too weak from his cold to rip it open. She wanted him to sneeze it down.
